This is one of 14 colorectal carcinoma cell lines derived by J.G. Park and associates during the years 1982 through 1985. The line was obtained from a fourth passage nude mouse xenograft of tumor fragments obtained from a patient prior to therapy (see also ATCC CCL-250.1 which was derived from the third nude mouse passage). |

Karyotype |
modal number = 46; range = 45 to 48. This is a pseudodiploid human cell line. The modal chromosome number is 46, occurring at 70% with polyploidy at 16.4%. The del (3) (p23), t(4,?), t(8;?) (p22), del (9) (p13), del (11) (p13), t(1;8) (p13;p24), t(1;?) (q11;?) and 2 other marker chromosomes are common to all metaphases examined. Other structurally altered chromosomes, if any, occur only sporadically. The t(1;8) and t(1;?) are not found in CCL-250.1. Normal N8 and N11 are absent. The X chromosomes are two copies per cell. The Y chromosome is not detected by Q-banding. |

Complete Growth Medium |
The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, ATCC 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (ATCC 30-2020) to a final concentration of 10%.

Subculturing |
- Remove and discard culture medium.
- Briefly rinse the cell layer with 0.25% (w/v) Trypsin- 0.53 mM EDTA solution to remove all traces of serum that contains trypsin inhibitor.
- Add 2.0 to 3.0 mL of Trypsin-EDTA solution to flask and observe cells under an inverted microscope until cell layer is dispersed (usually within 5 to 15 minutes).
- Note: To avoid clumping do not agitate the cells by hitting or shaking the flask while waiting for the cells to detach. Cells that are difficult to detach may be placed at 37°C to facilitate dispersal.
- Add 6.0 to 8.0 mL of complete growth medium and aspirate cells by gently pipetting.
- Add appropriate aliquots of the cell suspension to new culture vessels.
- Incubate cultures at 37°C.
Subcultivation Ratio: A subcultivation ratio of 1:3 to 1:6 is recommended
