Karyotype |
modal number = 46; range = 44 to 48. This is a pseudodiploid human cell line. The modal chromosome number is 46, occurring at 86%, with polyploidy at 16.2%. The ins(1;?)(p13;?), del(3)(p23), t(4q,?), t(8;?), del(9)(p13), del(11)(p13), 14q+ and two other marker chromosomes are common to all metaphases examined. Other altered chromosomes, if any, occur only sporadically. Normal N11 is absent. Two copies of X chromosomes are found in each cell. The Y chromosome is not found in QM stained preparations. |

This is one of 14 colorectal carcinoma cell lines derived by J.G. Park and associates during the years 1982 through 1985. The line was obtained from a third passage nude mouse xenograft of tumor fragments obtained from a patient prior to therapy. The line was initially grown in RPMI 1640 medium with 20% fetal bovine serum, but was later adapted to growth in the chemically defined medium ACL-4 (it can also be grown in medium with 10% serum). The cells and tumors formed from the cells produce detectable amounts of Dopa decarboxylase and the TAG-72 antigen. |
Complete Growth Medium |
ACL-4 medium supplemented with 10% FBS The base medium for this cell line is ATCC-formulated DMEM: F12 Medium Catalog No. 30-2006.
To make the complete growth medium, add the following components to the base medium:
- 0.02 mg/ml insulin
- 0.01 mg/ml transferrin
- 25 nM sodium selenite
- 50 nM Hydrocortisone
- 1 ng/ml Epidermal Growth Factor (do not filter)
- 0.01 mM ethanolamine
- 0.01 mM phosphorylethanolamine
- 100 pM triiodothyronine
- 0.5% (w/v) bovine serum albumin
- 10 mM HEPES
- 0.5 mM sodium pyruvate
- 2mM L-glutamine(for final conc. of 4.5 mM)
- 10% FBS

Subculturing |
Protocol: Cultures can be maintained by scraping off the attached cells and transfer along with the the floating cells into new flasks.
Medium Renewal: Two times weekly
Note: Growth is slow. |
