Animals were immunized with an acid soluble extract of rat (Sprague-Dawley) intermediate lobes.
Spleen cells were fused with Sp2/0-Ag14 myeloma cells.

Complete Growth Medium The base medium for this cell line is ATCC-formulated Dulbecco's Modified Eagle's Medium, Catalog No. 30-2002. To make the complete growth medium, add the following components to the base medium:
  • heat-inactivated fetal bovine serum to a final concentration of 10%
    • Subculturing Cultures can be maintained by addition of fresh medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 2 x 105 viable cells/ml. Maintain cultures at a cell concentration between 1 x 105 and 1 x 106 cells/ml.

    Medium Renewal: Two to three times weekly
    Cryopreservation

    Complete growth medium described above supplemented with 5% (v/v) DMSO.  Cell culture tested DMSO is available as ATCC Catalog No. 4-X.
